Analysis of viral and defective-interfering nucleocapsids in acute and persistent infection by rhabdoviruses.

چکیده
We have isolated and characterized the RNA of intracellular virus nucleocapsids recovered from a number of cell cultures persistently infected with rabies virus or vesicular stomatitis virus (VSV). VSV persistent infections in BHK21, L cells and Aedes albopictus (mosquito) cells generally showed the presence of large amounts of defective-interfering (DI) nucleocapsid RNA and much smaller amounts of standard (B) nucleocapsid RNA. Persistent infections of BHK21 cells by two rabies virus strains, challenge virus standard (CVS-11) or HEP-Flury, were followed for several months during which time the ratio of DI to B nucleocapsid RNA cycled dramatically. We also observed coordinated fluctuations in the absolute amount of incorporation of [3H]uridine into virus nucleocapsid RNA. Total incorporation was generally highest following a decrease in the relative amount of DI nucleocapsid RNA synthesis. At no time were DI nucleocapsids absent in any of the persistently infected cultures.

Prevalence of Persistent Bovine Viral Diarrhea Infection in Industrial Dairies of Tehran and Alborz Province, Iran
Background and Aims: Bovine viral diarrhea virus (BVDV) is considered as worldwide and economically significant infection. The aim of this study was to estimate prevalence of persistent bovine viral diarrhea infection (PI) in industrial dairies of Tehran and Alborz province, Iran. Materials and Methods: In this cross-sectional study, 102371 blood samples were collected from bovine of industria...
متن کاملIn vitro assembly of a functional nucleocapsid from the negative-stranded genome RNA of a defective interfering particle of vesicular stomatitis virus.
The template for transcription and replication of negative-stranded RNA viruses is a ribonucleoprotein structure, the nucleocapsid. We have developed a system that supports assembly of the negative-stranded RNA genome of a defective interfering (DI) particle of vesicular stomatitis virus (VSV) into a nucleocapsid in vitro. This system uses extracts from wild-type VSV-infected cells as a source ...
متن کاملPersistent bovine viral diarrhea virus (BVDV) infection in cattle herds
Bovine viral diarrhea virus (BVDV) is a significant pathogen associated with gastrointestinal, respiratory, and reproductive diseases of cattle worldwide. It causes continuous economic losses to the cattle industry primarily due to decreased reproductive performance. The ability of virus to cross the placenta during early pregnancy can result in the birth of persistently infected (PI) calves. P...
متن کاملInterfering With Lipid Raft Association: A Mechanism to Control Influenza Virus Infection By Sambucus Nigra
Sambucus nigra (elder) are broadly used species to treat microbial infections. Thepotential antiviral activity and mechanism action of elder fruit (EF) in human epithelium cell(A549) cultures infected with H9N2 influenza virus were determined. The effect of variousconcentrations of EF on influenza virus replication was examined by using virus titration, quantitative real time RT-PCR, fusion and ...
متن کاملInhibition of immunologic injury of cultured cells infected with lymphocytic choriomeningitis virus: role of defective interfering virus in regulating viral antigenic expression
The expression of viral antigens on the surfaces of lymphocytic choriomeningitis virus (LCMV)-infected L-929 cells peaked 2-4 days postinfection and thereafter precipitously declined. Little or no viral antigen was expressed on the plasma membrane surfaces of persistently infected cells, but LCMV antigens were clearly present in the cytoplasms of most of those cells. Cells early after acute inf...
